CORE FRAMEWORK : System State Model

The System State model defines physiology as a dynamic configuration of interacting domains—including metabolic, inflammatory, redox, and structural processes—that together determine how the body responds to internal and external inputs.
In this model:
• the same intervention may produce different outcomes depending on state
• variability in response is expected, not anomalous
• effective intervention depends on system readiness, not protocol alone

This framework provides the main conceptual foundation for all work developed within Clinicae.

ARCHITECTURE

Type: Computational architecture

OPAL translates the System State model into a structured, operational system. It organizes physiology into interacting domains and applies rule-based logic to evaluate:
• current system configuration • constraints on response
• appropriate timing and sequencing of interventions including energy production, redox, clearance ability, inflammatory response, and metabolism

OPAL is designed to move from static interpretation of data to state-aware decision logic.

DOMAIN MODELS (WITHIN MyOPAL): specific physiological systems are developed as domain modules.

These modules apply state-based logic to particular areas of biology.

EstroSafe — Estrogen System Modeling.
A model of estrogen physiology that evaluates how signaling, metabolism, and tissue response are shaped by system state with the goal of determining safe estrogen protocols for the individual woman.

Focus areas include:
• metabolite handling
• clearance capacity
• amplification context
• tissue vulnerability

OsteoMap — Bone System Modeling

A model of bone as a dynamic system influenced by energy availability, signaling balance, and remodeling capacity with the goal of helping each woman manage bone health based on her own body.

It integrates:
• genetic predisposition (via Stanford lab)
• metabolic + hormonal inputs
• structural + turnover dynamics


SYSTEMS MODEL: Drive Capacity Model

Type: Applied functional model

Drive Capacity Gap (published by Oxford University Press) is a functional expression of the System State framework. It describes how mismatch between:
• biological signaling (drive)
• system capability (capacity)
• and tissue-level execution (access)
leads to altered or incomplete physiological outcomes. It provides a practical lens for understanding how state manifests as lived function.
